Possible issues..

1) are you at the 1st frame of the video and have 'extend clips during transition' set to on? if so turn off or make sure you are not on frame 1

2)keyer not empty.. click inside the keyer layer (bottom most) and hit the delete key on your keyboard then try adding a transition.

Hi Mattie,

Thanks for your help.

No that's not issue.
I have tried just for you :-) but it doesn't work.

I just opened new project and I created two clips from one hour source clip.
Place them on timeline and tried to make transition between. It doesn't work.

can you place 2 color matte or color bar or still images next to each other and add a transition?
